What is 5-Amino-1MQ?
5-Amino-1-methylquinolinium (5-Amino-1MQ) is a cell-permeable small-molecule inhibitor of nicotinamide N-methyltransferase (NNMT), not a peptide. It is supplied as a lyophilized powder for in-vitro metabolic research.
NNMT is highly expressed in adipose tissue and liver, and its inhibition is a well-documented approach for probing the NAD+ salvage pathway in cell culture.
Mechanism of action
NNMT consumes S-adenosylmethionine to methylate nicotinamide, diverting nicotinamide away from NAD+ resynthesis. Inhibiting NNMT with 5-Amino-1MQ raises intracellular nicotinamide and, in adipocyte models, has been reported to increase NAD+ and sirtuin-1 activity.
Because the readouts overlap with NAD+ precursor work, 5-Amino-1MQ is frequently studied in parallel with NAD+ itself to distinguish salvage-pathway effects from direct cofactor supplementation.
Laboratory handling and storage
5-Amino-1MQ ships as a lyophilized powder in a sealed, tamper-evident vial. Store the unopened vial between −20°C and −80°C, protected from light, until the day of use. In this state the peptide is stable through the labeled expiration date printed on the lot documentation.
Allow the vial to reach room temperature before opening so atmospheric moisture does not condense onto the lyophilized cake. Introduce bacteriostatic or sterile water slowly against the vial wall, then swirl — never shake — until the solution is clear. Store the reconstituted solution at 2–8°C, aliquot to avoid repeated freeze–thaw cycles, and record the lot number in your laboratory notebook alongside the reconstitution date.
