Why Properly Lyophilized Peptides Don't Need Cold-Chain Shipping

Lyophilized peptides are shelf-stable for 18–24 months at room temperature. Cold-chain shipping is only required for reconstituted product.

The short version

A properly lyophilized peptide — sealed vial, moisture content under 5%, no reconstitution — is stable at room temperature for 18–24 months. Cold-chain shipping is required only for reconstituted (aqueous) peptide solutions, which degrade far faster.

Why lyophilization protects the peptide

Peptide degradation in solution is driven by water: hydrolysis of amide bonds, oxidation of methionine and tryptophan side chains, deamidation of asparagine and glutamine, and diketopiperazine formation at the N-terminus. Remove the water and most of these pathways slow by orders of magnitude.

A well-lyophilized peptide is:

  • <5% residual moisture (Karl Fischer titration).
  • Amorphous solid, not crystalline.
  • Sealed under vacuum or inert gas in the vial.

In that state, chemical stability is limited mostly by trace oxygen and light — both easily controlled with amber glass and a good stopper.

Published stability data

Multiple manufacturer stability studies and academic characterizations show:

  • 24-month room-temperature stability for typical lyo peptides.
  • 36+ months when stored at 2–8 °C.
  • 60+ months at –20 °C.

Most vendors label 18–24 months at room temperature as a conservative shelf life.

When cold-chain matters

  • Reconstituted peptides. Once you add bacteriostatic water, the peptide is in solution — refrigerate.
  • Peptides with oxidation-prone residues (Met, Cys, Trp) if the vial isn't sealed under inert gas. Cold storage buys margin.
  • Long-term storage beyond the labeled shelf life. –20 °C keeps material usable for years.

What under-drying looks like

If a vendor requires overnight ice packs on lyo product, one of two things is true: the product isn't actually well-lyophilized (moisture too high), or the vendor is over-cautious and passing the shipping cost on to you. Ask for the Karl Fischer moisture value. If it's under 5% and the vial is properly sealed, ice packs are unnecessary.
